Abstract


The purpose of this study was to isolate DNA probe which could identify Actinobacillus actinomycetemcomitans (A. actinomycetemcomitans) in Southern blot analysis. This study procedure includes (1) whole-genomic DNA extraction of A. actinomycetemcomitans ATCC 43717, A. actinomycetemcomitans ATCC 43718 and A. actinomycetemcomitans ATCC 33277 (2) construction of the genomic DNA library, (3) screening of restrictio fragment of genomid DNA, (4) identification of species-specific DNA probe by Southern blot hybridization. About 170 restriction fragments of several A. actinomycetemcomitans genomic DNA digested with the Hind III were obtained. One of them, the probe Aac6, restriction fragments of A. actinomycetemcomitans ATCC 33277, was shown to be hybridized with A. actinomycetemcomitans ATCC 43717, and A. actinomycetemcomitans ATCC 33277 but not A. actinomycetemcomitans ATCC 43728. From that result, the probe Aac6 may be useful in differentiation of A. actinomycetemcomitans ATCC 43717 and A. actinomycetemcomitans ATCC 33277 from A. actinomycetemcomitans ATCC 43178.
